Yiyue Xu works as a technician in the School of Life Sciences at Chinese University of Hong Kong (CUHK), specialising in the application and maintenance of cryo - EM, cryo - FIB - SEM, and cryo - CLEM equipment. The facility supports both single -particle analysis and cryo - electron tomography across diverse biological specimens, ranging from proteins to cells and tissues. Depending on sample size, specimens are prepared via plunge freezing or high-pressure freezing. An integrated Volta phase plate enhances image contrast, particularly for cryo - ET. Furthermore, lamellae are freshly prepared using cryo - FIB-SEM with regions of interest localised via fluorescent cryo - LM, while room-temperature, large-volume imaging is also supported.
